Abstract

Tumor microenvironment is the key of immune response regulation in cancer formation, which frequently causes dysfunction of CD8 1 T cells. Metabolic byproducts by tumor and stromal cells increases causes the metabolic balance of tumor-infiltrating lymphocytes. Lipid peroxidation is one of the key products. These compounds disrupt essential metabolic functions required, cytotoxic functions, protein stability, and mitochondrial performances. The long-term interference of metabolism exhaustion of T-cells in terms of lack proliferation, decreased cytokine secretion, and impaired anti-tumor immunity. The review provides existing knowledge about the function of lipid peroxidation and byproducts in the TME to the pathogenesis dysfunction metabolic perspective. It also describes the main molecular mechanisms of immune suppression caused by oxidative stress and discusses the possible therapeutic methods to reverse metabolism and effort the efficacy to the cancerous.
